Three-dimensional cultures of mucosal epithelial cells termed organoids are considered the most physiological models of primary epithelial cells in vitro and have a large number of potential applications in mucosal immunology research. Organoids can be generated either from pluripotent stem cells or from adult mucosal tissues. This protocol by McCracken et al. describes in detail how to derive human intestinal organoids from human pluripotent stem cells.
