Dendritic cells (DCs) play an important role in the maintenance of mucosal homeostasis and the induction of antigen-specific mucosal immune responses. DCs develop from bone marrow from increasingly restricted but phenotypically well-defined progenitor cells. This protocol from Michel Nussenzweig’s lab provides flow cytometry antibody panels and gating strategies that allow the simultaneous assessment of all human DC lineage–committed progenitors for identification, characterization or cell sorting.
